Mutai finished the 42.195km race with a time of 2:8:23 ahead of Ethiopia's Tsegaye Kebede (2:09:15) and South Africa's Lusapho April (2:09:45) in second and third place respectively.
Jeptoo clocked 2:25:07 to win the ladies marathon.
Uganda's double gold medalist Stephen Kiprotich did not impress this time after he ended the race in 12th position at 2:13:05. Kiprotich won the 2012 London Olympic Gold Medal as well as the 2013 Moscow Marathon gold medal.
Jackson Kiprop, also from Uganda featured in the top ten after he ended the race in 7th place at 2:10:56.
Despite showing interest in the top slot, Kiprotich lost his energy at the 34km mark when Mutai and Stanley Biwot, all of Kenya broke off.
